Environmentálna a potravinová mikrobiológia

Vedecká činnosť Laboratória environmentálnej a potravinovej mikrobiológie je zameraná na štúdium mikrobiálnych komunít zodpovedných za deterioráciu nášho kultúrneho dedičstva (nástenné maľby, drevené objekty, papier, pergamen, fotografie, textiílie, múmie, katakomby) a kvalitu tradičných slovenských potravín (syry, bryndza, slovenské vína) a tiež mnohých environmentálnych vzoriek (rastliny, vzorky zo vzduchu, pôdy, vody, metanogénna komunita bioreaktorov a syntetické polyméry) pomocou kultivačne závislých a kultivačne nezávislých metód. Okrem toho boli vyvinuté a optimalizované rôzne selekčné metódy na zoskupenie izolovaných mikroorganizmov. Oblasť vedecko-výskumnej činnosti laboratória smeruje k postupnému zavádzaniu molekulárno-biologických prístupov aj do spoločensko-vedných oblastí ako sú archívnictvo, knižné vedy, múzejníctvo, potravinárstvo a vinárstvo.

Laboratórium spolupracuje aj s ďalšími výskumnými skupinami na Slovensku, či už na pôde ústavov SAV (Ústav polymérov SAV, Ústav materiálov a mechaniky strojov SAV, Ústav ekológie lesa SAV, Ústav hudobnej vedy SAV, Ústav experimentálnej onkológie SAV) alebo v spolupráci s inými pracoviskami (Výskumný ústav potravinársky; Múzeum Betliar; Slovenská národná galéria; Slovenský národný archív; Slovenská národná knižica; Univerzita v Messine, Taliansko; Viedenská Univerzita, Rakúsko; Istituto Centrale per il Restauro e la Conservazione del Patrimonio Archivistico e Librario, Rím, Taliansko; Institute of Fermentation Technology and Microbiology at Lodz University of Technology; Department of Chemical Technology of Monument Conservation at University of Chemistry and Technology, Prague, National Cheng Kung University; Dep. of Environmental Engineering. Tainan City, Taiwan).

 Kolektív pracovníkov laboratória získal v roku 2016 cenu SAV za výsledky vedeckovýskumnej práce Deteriorácia kultúrneho dedičstva a kvalita tradicných slovenských potravín: štúdium ich mikrobiálnych komunít.

Fylogenomická ekológia

Výskum nášho laboratória je zameraný hlavne na štúdium genomiky, expresie, štruktúry a funkcie štyroch superrodín hémových peroxidáz, jednej rodiny hémových kataláz a jednej rodiny mangánových kataláz s možnosťou využitia vybraných antioxidantov v biotechnológiách.

Hémové peroxidázy a katalázy sú esenciálne enzýmové antioxidanty, prítomné vo všetkých formách života, aj v bunkách anaeróbov. Ich systematickú klasifikáciu možno nájsť na  http://peroxibase.toulouse.inra.fr/. Umožňujú odstraňovať reaktívne zlúčeniny kyslíka ktoré pri vyšších koncentráciách môžu poškodtovať všetky základné stavebné zložky bunky. Katalázy dokonca produkujú kyslík podľa rovnice: 2H2O2=2H2O+O2. Zameriavame sa na štúdium paralógov peroxidáz a kataláz u mikroorganizmov. Zverejnenie genómu pôdnej huby Chaetomium cochliodes naším laboratóriom v spolupráci s BOKU univerzitou (BMC Genomics 2016 - zoznam publikácií) ukazuje typický príklad distribúcie aj expresie génov pre peroxidázy a katalázy v genóme jednobunkového pôdneho eukaryota. Okrem toho je náš výskum zameraný tiež na štúdium obranného mechanizmu nesporulujúcich baktérií (Escherichia coli - modelový organizmus) voči rôznym stresovým faktorom prostredia (hladovanie, zmena teploty, oxidačný a kadmiový stres) a na štúdium diverzity bakteriálnych spoločenstiev a genetickej variability determinantov rezistencie proti ťažkým kovom v prostredí znečistenom rôznymi toxickými kovmi. Naše pracovisko je vybavené aj zariadením pre prácu s biologickými faktormi klasifikovanými ako faktory 2. skupiny.
